【摘要】:ThecompoundsthatbindtotheAh-receptorandinduceCYP1Ahaveoftenbeenshowntobeeithermoretoxicorcarcinogenic,orboth,thanotherorganiccompounds.Dioxins,furans,andPCBshavebeenshowntobioconcentrateinexposedorganismsandbiomagnifyinthefoodweb(seeGuideE1023).Testingwithbirds,mammals,andfishspecieshasshownthatexposuretothesecompoundscanproducephysiological,reproductiveandhistopathologicaleffects(12,13).Concernforthepossiblecontaminationofwater,food,wildlife,soil,andaquaticsedimentfromthesecompoundshasledtotherequirementforanalyticalchemicalanalysesofagreatmanyenvironmentalsamples.UseofascreeningtoolsuchasthisHumanReporterGeneSystem(HRGS)willallowidentificationofsignificantlycontaminatedsamples.Thesemethodswillaidinthecost-effectiveseparationofhighprioritysamplesfromthosethatdonotrequirefurthercostlychemicalcharacterization.1.1ThisguidecoverstherecommendedguidelinesforperformingatestforpresenceoforganiccompoundsthatbindtotheAhReceptorandinducetheCYP1Alocusonthevertebratechromosome.Underappropriatetestconditions,inductionofCYP1Aisevidencethatthecellshavebeenexposedtooneormoreofthesexenobioticorganiccompoundsthatincludedioxins,furans,coplanarPCBs,andseveralpolycyclicaromatichydrocarbons(PAHs).DetectionofinductionhasbeenmadesimpleandrapidbythestableintegrationofthefireflyplasmidsuchthatAh-receptorbindingresultsintheproductionofluciferase.Luciferaseproductionisafunctionofboththepotencyofthecompound(s)andtheconcentration.ThistypeofHumanReporterGeneSystem(HRGS)hasshownconcentration-responserelationshipsusing2,3,7,8-TCDD,5coplanarPCBs,andseveralpolycyclicaromatichydrocarbons(PAHs)(1-3).Thisguidedescribestestconditionsunderwhichsolventextractsofenvironmentalsamples(water,tissue,soil,orsediments)maybetestedforthepresenceofCYP1A-inducingorganiccompounds.1.2Thetestprocedurespresentedinthisguidehavebeenpublishedpreviously(4,5)andestablishedasEPAMethod4425(6).Thesereferencesshouldbeconsultedtoobtaindetailsregardingtheconstructionandmaintenanceofthecellline,andtheresponseofthecellstovariousorganicsubstances.1.3Alllaboratoryhealthandsafetyproceduresshouldbefollowed.Thisincludestheuseofglasses,gloves,andotherprotectiveclothing,whenhandlingthereagents.Informationontoxicity,handlingproceduresandwasteproceduresshouldbereviewedpriortouseofallchemicals.1.4Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
